Quantifying “Cliffs” in Design Space

چکیده

Purpose: This paper studies the regions of parameter space of engineering design in which performance is sensitive to design parameters. Some of these parameters (for example, the dimensions and compositions of components) constitute the design, but others are intrinsic properties of materials or Nature. The paper is concerned with narrow regions of parameter space, “cliffs”, in which performance (some measure of the final state of a system, such as ignition or non-ignition of a flammable gas, or failure or non-failure of a ductile material subject to tension) is a sensitive function of the parameters. In these regions performance is also sensitive to uncertainties in the parameters. This is particularly important for intrinsically indeterminate systems, those whose performance is ∗email:[email protected], Tel: 314-935-6202, Facs: 314-935-6219

Activity cliffs in drug discovery: Dr Jekyll or Mr Hyde?

The impact activity cliffs have on drug discovery is double-edged. For instance, whereas medicinal chemists can take advantage of regions in chemical space rich in activity cliffs, QSAR practitioners need to escape from such regions. The influence of activity cliffs in medicinal chemistry applications is extensively documented. However, the 'dark side' of activity cliffs (i.e. their detrimental...

Quantifying ice cliff evolution with multi-temporal point clouds on the debris-covered Khumbu Glacier, Nepal

Measurements of glacier ice cliff evolution are sparse, but where they do exist, they indicate that such areas of exposed ice contribute a disproportionate amount of melt to the glacier ablation budget. We used Structure from Motion photogrammetry with Multi-View Stereo to derive 3-D point clouds for nine ice cliffs on Khumbu Glacier, Nepal (in November 2015, May 2016 and October 2016). By diff...

On the validity versus utility of activity landscapes: are all activity cliffs statistically significant?

BACKGROUND Most work on the topic of activity landscapes has focused on their quantitative description and visual representation, with the aim of aiding navigation of SAR. Recent developments have addressed applications such as quantifying the proportion of activity cliffs, investigating the predictive abilities of activity landscape methods and so on. However, all these publications have worke...

Computational Methods Generating High-Resolution Views of Complex Structure-Activity Relationships

The analysis of structure-activity relationships (SARs) of small bioactive compounds is a central task in medicinal chemistry and pharmaceutical research. The study of SARs is in principle not limited to computational methods, however, as data sets rapidly grow in size, advanced computational approaches become indispensable for SAR analysis. Activity landscapes are one of the preferred and wide...

From activity cliffs to target-specific scoring models and pharmacophore hypotheses.

The role of activity cliffs in drug discovery projects is certainly two-edged: on the one hand, they often lead to the failure of QSAR modeling techniques; on the other, they are highly valuable for identifying key aspects of SARs. In the presence of activity cliffs the results of purely ligand-based QSAR approaches often remain puzzling, and the resulting models have limited predictive power. ...
